Step by Step

How to reconstitute peptides

Reconstitution turns a lyophilised freeze-dried peptide powder into a measured liquid solution ready for research use. Get this right and the rest of the protocol is straightforward.

What you need

  • Bacteriostatic water with 0.9% benzyl alcohol
  • An insulin syringe (29G, 0.5mL or 1mL)
  • Alcohol prep pads
  • Your lyophilised peptide vial
  • A clean flat work surface

Step 1. Prepare the environment

Wash hands thoroughly and wear clean non-sterile gloves. Wipe the work surface with 70% isopropyl alcohol and lay out every item you need before starting.

Step 2. Calculate the volume

Use the calculator above. Divide the vial strength in mg by the water volume in mL for concentration, then divide your target dose by that concentration for the volume per dose. On a 1mL insulin syringe, 1mL is 100 units.

Step 3. Add the solvent

Wipe the vial rubber stopper with an alcohol swab and let it dry. Draw the calculated volume of bacteriostatic water into the syringe. Inject the water slowly down the inside wall of the peptide vial rather than straight onto the powder.

Step 4. Mix gently

Do not shake. Gently swirl or roll the vial between your palms until the powder is fully dissolved. The solution should be clear and colourless. Cloudiness or particles mean something is wrong, so stop and inspect.

Step 5. Store correctly

Refrigerate at 2-8C immediately after reconstitution. Keep away from direct light and avoid repeated freeze thaw cycles. Most reconstituted peptides stay stable for 28-30 days refrigerated. Use a new sterile needle and syringe for every withdrawal.
